Most scintillating of all, some supremely talented stars sing their bloody hearts out. Hugh Jackman mesmerizes as Jean Valjean, an ex-prisoner who breaks parole in 1815 France, then gets hunted for the next 17 years by his prison guard, Javert (Russell Crowe).

Along the way, Valjean encounters ailing factory worker Fantine (Anne Hathaway) and decides to raise her daughter (Amanda Seyfried in adulthood). The actors eschew the lip-synching and go live (often in extreme close-up), which heightens the drama: During a confrontation between Valjean and Javert, the baritone Crowe sings with such anger, the spittle ricochets onto Jackman's face.

Newcomer Samantha Barks, as heartsick Eponine, openly weeps during "On My Own." And, in limited screen time, a broken-down Hathaway leaves an indelible impression, crushing "I Dreamed a Dream."

Stop cringing, musical haters!

A film that oozes this kind of passion only works at full tilt. So embrace it and, yes, hear the people sing.

Chinese professional social network Ushi launches ... - The Next Web

Ushi, often referred to as the Chinese LinkedIn, has released an app for scanning and storing name card information, as it works to build momentum for its professional social network.

The first thing you do in just about any Chinese professional setting is whip out a name card. Lots of cultures put the time-honored practice to use, but it seems to me that the East Asia region is the most strict in its observance of business cards and introductions. For countries like China, Korea and Japan where the concept of ?face? is a crucial part of doing business, name cards and the titles and roles they convey are essential.

But they?re also antiquated. We carry paper around with us, despite the face that we have powerful mobile computers at our fingertips. Often those business cards end up in a drawer collecting dust and we?re unable to find the one we need when it?s time to reconnect.

Several startups are tackling the problem. Yolu Card Reader and Evernote?s Yinxiang Biji Renmai (the localized version of Evernote Hello) come to mind, but Shanghai-based Ushi, as the latest entrant, has the weight of its social network behind it.

With 850,000 users, Ushi isn?t huge. It started out as an exclusive invite-only network targeting executives and entrepreneurs, though it has since opened up registration to all. Ushi Mingpian (Ushi Name Card) is designed to continue driving user growth by solving the name card problem. The app is currently available for both iPhone and Android.

Ushi CEO and co-founder Dominic Penaloza believes the app will help new users to a real-name professional social network because they?re already familiar with the concept of a name card being an important real-name tool. Whether Chinese netizens like it or not, real-name requirements are going to be more and more common on the country?s Internet, if not fully mandated.

?When two business people exchange business cards, not only is it the fundamental first thing they ever do, but it helps us to explain a lot of things. The name card of course is real-name. It?s for business, that?s what it?s for,? Penaloza said.

The foundation of the app is a name card scanner, but it?s more importantly designed to be a self-updating address book.

?I think we?re in the best position in the market to deliver on the promise of the self-updating address book and create even more value for users,? he said, adding that the app balances the utility of card scanning with social aspects.

In its 2 1/2 year history, Ushi has managed to attract a highly valuable demographic to its professional network. The average age is 31 years old, so it skews heavily toward middle-upper management. One-third of Ushi?s users are in the technology, media and telecom industries, while 20 percent are in professional services. Penaloza also estimates there are 15,000 headhunters, 25,000 HR professionals and about 7,000 venture capitalists on the network.

The company?s exclusive beginnings helped to Ushi to court an older crowd. Prior to launch, the team spent months in face-to-face meetings with Chinese CEOs and business leaders to convince them to become charter members. Penaloza says the network grew to 50,000 members in five months, roughly the same speed as Facebook. A year later, it was at 300,000 members and the company decided to open up registration to speed growth.

A number of prominent Chinese angel investors have backed Ushi, as well as one of its clients, expert network Gerson Lehrman Group. In April, Ushi began offering commercial services to source experts for GLG.

?It really works. It?s becoming one of [GLG's] efficient channels,? said Penaloza.

Ushi says it doesn?t need to directly make money from the new card-reading app. Penaloza compared his company?s status to that of LinkedIn, in that their revenue stream is robust enough not to have been adversely affected by the switch to mobile and the resulting lower revenue streams that non-professional Internet companies are dealing with.

?It gives us more liberty to make a great smartphone app and not worry directly about monetizing the users of it,? he said.

Tianji, China?s leading professional social network, expects the market to grow to 100 million?next year. If that?s the case, Ushi?s going to have to move fast to secure a decent share of it, since it has yet to top 1 million. The industry is heating up fast, as Sina recently revealed that it is spinning off a business-focused subset of its Weibo microblog. Ushi?s new app is a step in the right direction, so this is definitely going to be an interesting space to watch next year.

The Google Search app does some things better than Siri.

Apple promises a lot from Siri. The company advertises this "intelligent assistant" as a major selling point for new iPhones and iPads. Ask a question out loud, and Siri digs up an answer; dictate an appointment, and Apple's software will jot down the date for you. At least, that's the idea.

"It never un?derstands my voice," says Amanda Soloway, an iPhone owner and student in the University of Washington's Master of Business Administration program in Seattle. "I think there was one time that I got it to work. I got it to put something on my calendar. But by the time Siri figured out what I was saying, I could have just done it myself."

Ms. Soloway echoes the complaints of many unhappy Siri users. In specific cases, Apple's ?robot-voiced helper can be quite a crowd pleaser. (Ask it to "tell me a joke" or "what's '2001: A Space Odyssey' about?") But when it comes to legitimate utility, Siri is often lacking.

Things look even worse for Siri now that Google is trying to beat Apple at its own game. The iPhone's Google Search application recently got a big update that unlocks voice search. The free app works much like Siri. Press the microphone icon, say your query aloud, and Google attempts to sniff out an answer.

Both Google Search and Siri do well with questions like "What's five pounds in US dollars?" or "Who won the Cowboys game?" Apple and Google programmed their software specifically to handle common inquiries such as these.

However, if Siri doesn't know the answer to your question or can't figure out what you're saying, then Apple passes the buck. For example, ask Siri "Who painted the Mona Lisa?" and it will reply, "Would you like me to search the Web for 'Who painted the Mona Lisa'?" If you agree, then Siri hands you over to Google.

Why not just start with Google? In our tests, Google voice search delivered answers faster than Siri could, it had an easier time figuring out what we meant, and the Google Search app didn't require an awkward handoff for tough questions.

That said, Siri is built into Apple's products, so it can access parts of the device that Google simply can't. The Google Search app can't text-message friends. It can't access your calendar. It can't function in a hands-free mode. Siri can.

However, if you want quick answers to life's little mysteries, try out Google Search. It's available in the iTunes store.

Things You Want Know Before Building Wooden Garden Sheds ...

A wooden garden shed can provide you with many benefits. Many homeowners consider building their own garden shed to store garden tools, landscaping equipment and other supplies. Some will also use this building for a safe environment to establish seedlings before moving them outdoors. A garden shed may be larger or smaller in size, and it can add function and convenience alike to your gardening experience. Before you start building your own garden shed, however, there are a few things you should consider fully.

The Need for a Construction Permit
Many municipalities require homeowners to obtain a construction or building permit before adding a structure to their property. Depending on the size of your shed and where you live, this may not be required. However, if you build a shed in an area where a construction or building permit is required and without getting that required permit, the city or municipality could require you to remove it from your property. You may invest a lot of time, sweat and money into building your shed, so you do want to ensure your building plans comply with local laws.

The Type of Shed
If you only have plans to use your shed for storing lawn and gardening equipment and tools, a basic shed without insulation, electricity, water and more may be acceptable. You may not even need to install a window if the shed will be used for a basic storage purpose. However, if you are storing items that need to be kept at a steady temperature, insulation and possibly climate control capabilities may be required. If you will be using your shed to store extra large equipment, such as a riding lawn mower, you may need a double-wide door along with an entrance ramp. It may be necessary to have a pass-through experience with doors on opposite ends of the shed. If the shed will be used to raise seedlings, ample natural lighting with the installation of windows coupled with a sink for easier watering may be useful. You want your shed to conveniently meet all of your needs, so picking out the best shed plans possible is key.

The Location
Any time you build a new structure on your property, you want to consider the location carefully. This is even more true when building a wooden garden shed. Keep in mind that a structure will need to be placed in a relatively flat area without an incline. If you choose an area with an incline, this area will need to be leveled so that you can build on a flat surface. It should also be an area that is not prone to flooding or developing pools of standing water during wet times as a wooden shed could rot when exposed to significant water over time. Further, the contents inside the shed could be damaged.

The Cost
Many homeowners debate between building their own shed from scratch, often using do-it-yourself plans found online, and installing a pre-fabricated shed on their property. It can be difficult to fully estimate the cost of building a shed on your own from scratch. To start, you will need to find the right plans to utilize. The best plans include a list of all of the materials and tools you will need for the job. You can take this list to the local home improvement store, locate the items and calculate the cost. Keep in mind that any tools required will either need to be leased or purchased. In most cases, it is more affordable to build your own shed from plans found online.

Building your own wooden garden shed is not something that you want to undertake without diligent planning. It is wise to spend ample time finding the perfect location for the shed in your yard and researching the need to apply for a building permit. While you wait for your building permit to be approved, you can gather the needed supplies and materials for the project. Most sheds can be completed with a few days of diligent effort, but some may require you to work on them over the course of several weekends.

Thirty years later, a Jefferson Parish crime victim confronts and ...

"It's not that I wouldn't have wanted to grow up with an eye and a father, but it made me who I am today." -- David Lyons

One of David Lyons' last memories of the New Orleans area is waking up in a hospital room surrounded by stuffed animals and get well cards from people he didn't know. The walls were lined with plush playthings, including plenty of E.T.s and colorful Smurfs.

Family members from Colorado, whom Lyons had never met, also were there.

It was 1982, and the 5-year-old boy awoke - after three days - to some cold, harsh facts: robbers' bullets had destroyed his right eye and torn apart his small body, his father had been shot to death, and life as he knew it would never be the same.

After about 10 days of recuperation at Ochsner Foundation Hospital, relatives took Lyons back to Colorado, where their plane touched down in a fierce snowstorm. He never set foot in Louisiana again.

Now, 30 years after the horrific shooting that he occasionally refers to as "the accident," Lyons for the first time has been reaching out to learn more about what happened on that Halloween night in 1982 along Jefferson Highway in Jefferson. And he hopes to return to Louisiana some day.

"I never really pursued it," Lyons, who lives in Golden, Colo., with this girlfriend, said in a recent interview. "I left it alone, made peace with it and moved on with my life.

"But I always wanted to know a little more about what happened and look up some of the people that my father knew ... and get a little closure."

"More jewelry than Mr. T''

Two weeks after being born in Denver, Lyons and his family moved to the New Orleans area in 1977, where his father, Vincent, had visited once and long dreamed of settling. After three years here, Vincent and Vanessa Lyons divorced, and David remained with his father.

Vincent Lyons, of Leadville, Colo, was a flashy jeweler who operated a booth, apparently at the French Market in the French Quarter.

"He wore more jewelry than Mr. T," David Lyons said. "He was a showboat .... He would have $100s rolling out of his pockets."

On that fateful Halloween, David's father dressed him up as a devil and painted his face red before dropping him off at a babysitter's house. The sitter took Lyons and other children trick-or-treating that night before Vincent Lyons, 39, arrived in his green Oldsmobile Vista Cruiser station wagon to take his son home.

Lyons said he remembered standing up in the front passenger seat - he loved doing that - at about 8:30 p.m. when his dad pulled into Trailer Town at 4038 Jefferson Highway, where they lived. Suddenly, without warning, bullets began screaming into the car.

"That's when everything went Pow! Pow! Pow! Pow! Pow! out of nowhere.

"I remember it being hot. Extremely hot. In my eye and my stomach and my leg."

A .38-caliber bullet tore through Lyons' eye; two slugs ripped open his abdomen, with one exiting his back; another hit his leg. His father was shot multiple times and later died at Ochsner.

Lyons apparently blacked out, and he recalls being on the ground when he came to. He'll never forget the searing heat from the bullets that tore through his small body.

"Unbearable amounts of heat ... and at the same time I was freezing cold. I kept asking for a blanket. I was in shock.

"I remember playing with my guts. People kept pulling my hands away."

And then Lyons awoke in the hospital days later, his world shattered.

Jefferson Parish sheriff's deputies found a large amount of blood on the station wagon's driver's seat and passenger seat, as well as on the shell roadway outside the open driver's side door.

In typical fashion, Vincent Lyons was wearing and carrying a lot of jewelry on the day that he died.

Court records said the Jefferson Parish Coroner's office turned over to investigators the many items found on the elder Lyons' body: 11 gold rings, a gold wrist watch, a gold stick pin, gold money clip, two gold chain necklaces with attached charms, three plain gold necklaces, one large gold bracelet, four small gold bracelets, one gold "charm of the world," two gold cuff links, an "unknown animal tooth" and $583 in cash.

Three suitcases containing jewelry were found in the back of the station wagon.

Less than a month after the shooting, three men were booked with the crime. Juan Caceres, Rigoberta Machado and Libio Calzadilla were described by authorities as Cubans who came to the United States in the 1980 freedom flotilla in which thousands of Cubans fled their country in makeshift boats and floated to Florida.

About a year later - Nov. 30, 1983 - the charges against the trio were dismissed by the Jefferson Parish District Attorney's Office. The court record does not list a reason for the dismissal.

Growing up

A .38-caliber bullet still lodged in his head, David Lyons was raised in Denver by his paternal grandparents, Robert and Petra Lyons.

His father dead, his mother living in North Carolina, and sporting a glass eye, Lyons endured some tough times as a youngster. "Growing up was hard . . . kids can be cruel," he said.

When he was 7 or 8, he rummaged through his grandparents' closet and came across a Nov. 20, 1982, newspaper story from The Times-Picayune: "3 are booked with Halloween killing, shooting."

For another 27 years or so, that's about all Lyons would know about the crime that robbed him of his father and his eye.

At age 15, Lyons left school, left home and got a $5-per-hour job cleaning out cat boxes for an animal rescue outfit. "I was one of those geniuses who dropped out," he said of his decision to call his education quits in 10th grade.

He held a variety of jobs over the years, including operating the deli for a Wild Oats store. Lyons became a certified nursing assistant and worked in the home health, longterm care and hospice fields.

By 2002, while he was living in Phoenix, Ariz., the bullet in his head had worked itself down to his jaw. It was time for it to come out.

The flattened-out slug, which was too close to a nerve for doctors to remove in 1982, was taken out nearly 20 years to the day that it bore into Lyons' head. Arizona authorities kept it for about 60 days before releasing it to Lyons, who drilled a hole in it, slipped on a necklace and wore it around his neck.

"That piece of metal took my eye," he said. "I wasn't going to let 'em just throw it away."

Lyons eventually lost the trophy slug somewhere in Arizona, but isn't bothered by it. "I don't need it anymore."

Embracing the past

Lyons returned to Denver in 2006 and recently moved to nearby Golden, where he lives with his girlfriend of one year, Barbara Miller, and works for Spyderco, a knife manufacturing company.

It was Miller who began asking questions about that Halloween night 30 years ago. Without any prompting from Lyons, she started doing research and contacted The Times-Picayune to request whatever stories the newspaper had on file.

Three stories were emailed to Lyons on Halloween, the 30th anniversary of the shooting.

"Best Halloween ever," a thankful Lyons told a reporter after receiving the email.

"I pretty much did it myself,'' Miller said. "I started wondering. And I thought he needed to know."

"Once I started finding things, he was really glad and wanted to know more. It's definitely been good for him."

Lyons, now 35, said that while he never pursued the details of what happened, "I've always been on the road to find out. All these questions have always been with me."

He said he holds no ill will toward Louisiana and has forgiven the men responsible for the shooting.

In spite of it all, "I really think I turned out to be a decent young man. I have a kick-ass career, a big house and ... the greatest woman in the world. How could you ask for more than that?

"In some ways, you're thankful for what happened ... Losing an eye and a father at the same time really shapes a man. It really sets the path.

"It's not that I wouldn't have wanted to grow up with an eye and a father, but it made me who I am today."

HONOLULU (AP) ? The late Sen. Daniel Inouye was remembered Sunday as an American hero whose legacy as a war veteran and longtime senator would be felt across Hawaii for years to come.

The memorial service at Honolulu's National Memorial Cemetery of the Pacific was attended by more than 1,000 people, including President Barack Obama, Senate Majority Leader Harry Reid, Hawaii's congressional delegation and a number of other senators, cabinet secretaries and other dignitaries.

"Daniel was the best senator among us all," Reid told those assembled. "Whenever we needed a noble man to lean on, we turned to Sen. Dan Inouye. He was fearless."

The cemetery is the final resting place to thousands of World War II veterans. More than 400 members of the storied Japanese-American 442nd Regimental Combat Team ? of which Inouye was a part ? are buried at the site.

Several 442nd veterans attended the Sunday morning service, the latest in a number of tributes and honors for Inouye following the 88-year-old's Dec. 17 death from respiratory complications.

A 19-gun cannon salute was fired as Inouye's coffin arrived at the cemetery. The service also featured a flyover by F-22 military jets and the playing of "Taps" by Democratic Sen. Jon Tester of Montana.

Inouye's widow, who was seated with the president and first lady Michelle Obama in the front row, dabbed her eyes as a pipes and drums band played "Danny Boy."

Inouye was the first Japanese-American elected to both houses of Congress and the second-longest serving senator in U.S. history.

He was a high school senior in Honolulu on Dec. 7, 1941, when he watched dozens of Japanese planes fly toward Pearl Harbor and other Oahu military bases to begin a bombing that changed the course of world events.

He volunteered for a special U.S. Army unit of Japanese-Americans and lost his right arm in a battle with Germans in Italy. That scratched his dream of becoming a surgeon and he went to law school and into politics instead.

"He was a shining star of the greatest generation," fellow Hawaii Democratic Sen. Daniel Akaka said at the service.

Akaka also highlighted Inouye's role in steering federal money to build roads, schools and housing in Hawaii over the decades, from the beginning of statehood.

"Dan Inouye is Hawaii and Hawaii is Dan Inouye," Akaka said.

Several services have already been held in Washington and in Hawaii for Inouye. He lay in state at both the U.S. Capitol Rotunda on Thursday and the Hawaii state Capitol on Saturday.

Obama eulogized Inouye during a service at Washington's National Cathedral on Friday, saying that Inouye's presence during the Watergate hearings helped show him what could be possible in his own life.

The president arrived early Saturday in Honolulu for his annual Christmas family vacation. He made a brief visit to the grave of his grandfather, World War II veteran Stanley Dunham, after Sunday's service.

As I pulled the bundle of mail from our mailbox, I sorted through it. Sandwiched between the magazines and credit card offers was a Pepto Bismol pink colored envelope. I flipped it over and a sticker with smiling jungle animals made eye contact with me. I carefully opened it and pulled out a beautifully handcrafted invitation. It was edged in pink and Dr. Seuss bright green stripes. All the friendly animals of the jungle were jumping around the paper. I was expecting this baby shower invitation. Our friend is having her first baby. The baby is a little girl and she has already been named Olivia. This shower will be a celebration of life. A time of anticipation and excitement about the little one who will hopefully make her appearance before Christmas Day.


The shower was held at a very exclusive restaurant. I had been informed that a full luncheon would be served. As I drove to Cleveland, I pondered how many baby showers I had attended in my life. I recalled going to baby shower breakfasts, brunches, picnics, luncheons, after hour parties, during lunch breaks, and open houses just to name a few. I have watched as the Mommy-to-be gets feted with rocking chairs, umbrellas, bunny slippers, floating loungers, recliners, pillows, bubbles, chocolate molded baby buggies, lemonade with curly straws, 500 calorie petit four, neck massages, pedicures, manicures, balloons with Barbie's face, everything pink, everything blue, and everything Disney. I have witnessed every theme from swimming pools to Mother Goose to frogs.?
This baby shower was probably the fanciest I have ever attended. It was complete with the Father-to-be, the Grandparents-to-be, all the aunts and uncles-to-be. Everyone was invited. We were seated at linen clad round tables with crystal glasses, porcelain plates, and real stainless steel utensils. There were none of the baby pink and baby blue paper plates that I host with. Gone were the plastic forks wrapped in paper napkins and curly tied with rainbow ribbon. We feasted on a delicious meal. Prime Rib, chicken Parmesan, Italian pastas and fresh homemade bread. There was an enormous strawberry filled multi-layered cake and gourmet cookies and?gelato and other delicacies. Chocolate hand painted giraffe pretzel rods complimented each table setting. Someone had made "take home goodie bags" filled with more chocolate ...one for everyone in attendance. I felt pampered and I was only a GUEST! This family is greatly anticipating the birth of this little baby. They had planned and dreamed and worked on this baby shower for months. It was a one of a kind baby shower experience. The gift table trailed at least 20 feet long and was laden with every design of baby shower paper and gift bags. I received a lesson in gift wrap design and bow creation by just looking over Baby Olivia's presents. There were the trendy diaper cakes, multi colored and shaped teething rings attached to the handles of the gift bags, stuffed animals peeking out of packages, and boxes wrapped in receiving blankets instead of paper gift wrapping. The wrapping and creativity of many that bestowed gifts on this new Mother was beyond words. Any baby item that has been produced had been purchased for Baby Olivia.? Gift after gift was opened and shared for all to see. I was especially interested in gifts made of fabric. Little soft sleepers and Onesies. Delicate cotton and baby yarn layettes made by a loving Great-Grandmother. Downy blankets and hoodies in every color. Quilts made from quality 100% cotton?fabrics...all created for precious long lasting usage and for leaving a legacy. Every fabric gift was primed for sensitive baby skin. Fragile little baby fingers and toes that would peek out of the blankets. Silken first baby hair that would be hidden in those folds of fabric. Every gift was given from the heart and with love. Given with thoughts of making little Olivia feel welcome and cherished. It was wonderful watching the new parents with their gracious attitude of appreciation for each of their guests. They are a kind and caring couple that truly cherish their family and friends. Little Olivia will grow up well under the watchful care of these two wonderful people.

When I think about the atmosphere of anticipation at that baby shower, I can only imagine how Mary and Joseph must have felt. They prepared and planned for the coming Jesus. Joseph was a carpenter. His passion was building. The Bible doesn't say this... but?I imagine?that he had crafted a cradle of sorts for the dear baby to be lain in. It was probably the best one he had ever created. I can see in my mind's eye the labor of love. The rough strong manly hands on the warm olive wood. Sanding and smoothing, shaping that piece of wood into a work of art, making everything perfect for the infant, using his finish carpentry skills to design a unique creation. The pride of Joseph's skill went into the bed of a King. The personal sacrifice that turned the wood log into a masterpiece.? Mary was a servant girl. She had been trained to care for the home.? She had likely tended to family while she was but a child herself.? Every possible act of love was poured into the forthcoming birth of her baby. She would have?sought out the cleanest, purest, softest, and most durable fabrics.?She would have taken care of her body. No doubt in my mind, that she was very cautious where she went, what she ate, what she drank, and who knew about her precious pregnancy. Extreme care was foremost in her and Joseph minds as the days grew near.

Then the announcement...?Caesar's?sinister plan. Everyone was to return to the city of their ancestors... NOW!? No exceptions... DO IT NOW! Don't question... follow the rules... or else. What a shock to the young couple. They had been planning an easy and safe birth. Their families were close and would assist in the birthing. Now... they were thrust out into a cold uncaring world... just the two of them... and God. All their well made plans... cast aside.? They couldn't even take the beautiful olive wood cradle that Joseph had lovingly handcrafted for Baby Jesus.? They knew they had to follow the law and travel to Bethlehem. It would be a hard journey for a woman with child. There might not be any place to find refuge and safety.?They knew and yet they obeyed. There is not one mention of resistance in the Word of God. They did exactly what God told them to do.?


All was ready in Bethlehem the night that Jesus came to earth. Mary and Joseph were prepared. God had prepared more than just their hearts. God had prepared the nation of Israel, the town of Bethlehem, the star, the donkey that carried Mary, the inn keeper that offered his animal stable, Joseph's willingness to take Mary as his wife, and?Mary's first experience with giving birth to her own child. God had prepared every part of Jesus' birth. The people, the animals, the stable, and the nation were all anticipating His arrival. Mary was ready. Joseph was ready. The inn keeper was ready.?All was in place and part of God's magnificent plan. Jesus was sent from Heaven, nurtured in a virgin womb, protected by a son of David, carried by a humble donkey, born in a lowly stable, laid in a stone feeding trough, and wrapped in strips of soft cloth. Cloth that was prepared just for a newborn body. Cloth that was torn from fabric that would protect the tiny baby from the cold and evil world He had just entered.??God had put everything in place for Jesus' arrival on Earth. There was no rushing, no worrying, no questioning. It was the way God wanted it to be on that very first Christmas Day.?

Newly-discovered Cyclin D1b/hormone receptor pathways axis promotes aggressive forms of prostate cancer

PHILADELPHIAElevated levels of Cyclin D1b could function as a novel biomarker of lethal metastatic disease in prostate cancer patients, according to a pre-clinical study published ahead of print on December 21 in the Journal of Clinical Investigation by researchers at the Kimmel Cancer Center at Jefferson.

The group, headed by Karen E. Knudsen, Ph.D., Professor and Hilary Koprowski Chair, Departments of Cancer Biology, Urology, and Radiation Oncology at Thomas Jefferson University and Deputy Director for Basic Science at the KCC, found that Cyclin D1b, a variant of the cell cycle regulator Cyclin D1a, functions independently of the cell cycle to promote metastasis in both early and late stage prostate cancer.

Rather, Cyclin D1b, but not Cyclin D1a, regulates a large gene network, the researchers found, which was shown to cooperate with androgen receptor (AR) signaling to fuel metastatic progression in multiple models of prostate cancer.

Studies have shown that Cyclin D1b expression is elevated in early stages of prostate cancer (in up to 30% of primary disease), and researchers have now demonstrated that this occurs more frequently in late stage castration-resistant prostate cancer: up to 80%.

Cyclin D1b expression is also highly correlated with that of the pro-metastatic gene SNAI2 (Slug), which the group identified as regulated by cooperative signaling between Cyclin D1b and AR.

"Numerous clinical and pre-clinical studies have effectively demonstrated that AR signaling is critical for progression to metastatic disease, but our knowledge of AR targets which can induce metastatic phenotypes is limited," said Dr. Knudsen. "Our data describe how cross talk between the cell cycle and AR can rewire the AR signaling axis to enhance the expression of genes which elicit metastasis in both early and castration resistant prostate cancer models."

"We found that Cyclin D1b can directly promote AR dependent expression of the gene SNAI2 (Slug), which dramatically increased metastatic events to soft tissues in animal models," she added.

Metastatic castration resistant prostate cancer represents the most lethal form of the disease, which arises when AR is reactivated despite continued hormone therapy.

Soft tissue metastasis to the liver and lung represents a particularly aggressive form of prostate cancer, whose presence predicts for decreased survival time in prostate cancer patients.

Currently, there is little knowledge as to how these metastatic events occur, and identification of pathways and biomarkers of this lethal event could greatly benefit prostate cancer patients.

Using various in vitro and in vivo models, researchers found that Slug enhances the ability of cells to colonize soft tissues, which resulted in a higher incidence of metastasis in the liver and lung.

Given the inability to manage AR signaling in metastatic castration resistant prostate cancer, Slug driven pathways could be leveraged to dramatically limit the incidence of soft tissue metastasis and improve patient morbidity and mortality, researchers believe.

"Identification of AR driven pathways which mediate metastatic progression represents a significant leap forward in our attempts to effectively manage prostate cancer progression," said Dr. Knudsen. "Cyclin D1b and Slug can likely be used as biomarkers to identify patients with an increased risk of metastasis, and will eventually provide us with novel "druggable" targets downstream of AR and Slug which can be exploited to dramatically reduce the incidence of these lethal metastatic tumors."

Young scientist helps identify cause of widespread eye disease ...

Branch retinal vein occlusion ? blockage of the blood vessels that channel blood from the retina ? is a common eye disease. A type of blood clot in the eye, the disease causes reduced vision, and people with the disease also typically have an increased risk of hypertension, diabetes and other serious conditions. A young scientist from the University of Copenhagen has made a significant contribution to finding the cause of the disease.

A team of researchers at the University of Copenhagen, Glostrup Hospital and several other ophthalmology departments at Danish hospitals have now shown that it is highly probable that thickening of the arterial walls is behind the common eye disease known as branch retinal vein occlusion ? a type of blood clot in the eye that blocks the vessels that transport blood from the retina. The disease leads to reduced vision and affects more than 14 million people worldwide.

?Our new results indicate that branch retinal vein occlusion is caused by thickening of the arterial wall. This makes it crucial for doctors to treat patients diagnosed with the disease with medicine to lower blood pressure in order to prevent blood clots from forming in the heart and brain. Branch retinal vein occlusion is often a sign of increased risk of blood clots in other parts of the body,? explains Mette Bertelsen, PhD student at the University of Copenhagen.

Targeted treatment
Mette Bertelsen, head of the research project, and her colleagues photographically verified the diagnosis of branch retinal vein occlusion in 1168 people. They identified the patients? other diseases with the help of Danish national registries and compared the data to that of 116.800 healthy people.

By looking at the illness and mortality statistics of Danes diagnosed with a blood clot in the retina?s main blood vessels, both before and after the occurrence of the retinal blood clot, Mette Bertelsen and her colleges has now shown that while these patients show a higher frequency of arterial disease in the heart and brain, they do not display a higher frequency of venous disease. This new knowledge, which has been published in the British Medical Journal, means that disease prevention and treatment of these patients should be targeted at hypertension, diabetes and Atherosclerosis, while doctors can save patients from unnecessary treatment with anticoagulants.
Thickened arteries the villains

Doctors have long debated the three most likely theories about what causes branch retinal vein occlusion. Discussion centres on whether the disease is due to a) conditions in the veins that cause blood clots, b) a change in the composition of the blood, or c) blockage of the vein due to compression from an adjacent artery that has thickened and thus compresses the vein and hinders blood flow.

?To understand what is actually happening, it can be helpful to picture a garden hose that has been squeezed by a larger hose, cutting off the water supply. That is essentially what happens when a vein is compressed by a thickened artery. Clearly, the consequences can be serious,? explains Mette Bertelsen. She adds that no one knows exactly how many Danes suffer from branch retinal vein occlusion, but that more than 14 million people have the disease worldwide.

Apple chief executive officer Tim Cook talked extensively about management and corporate creativity in an interview with Bloomberg Businessweek editor Josh Tyrangiel.

Mr. Jobs was a major shareholder in the Walt Disney Company and had seen how executives there wasted time trying to figure out what Disney himself would have done after the founder of the company had passed away. Jobs ?removed a tremendous burden for me,? Cook says, by instructing, ?I never want you to ask what I would have done. Just do what is right.?

Apple has taken heat for poor working conditions at massive Foxconn Technology Group factories in China where many of its products are assembled. Cook told Businessweek that Apple would start producing one line of its Mac computers in the US in 2013 ? a modest sign of the brightening prospects for US manufacturing mentioned above.

Creativity, in Cook?s definition, is ?people who care enough to keep thinking about something until they find the simplest way to do it.? He laughed about corporate innovation departments saying that having one ?is always a sign that something is wrong ... you know, put a for sale sign on the door.?

Authorities: Kansas man who killed 2 cops dies

TOPEKA, Kan. (AP) ? A man with a history of theft and weapons convictions gunned down two police officers investigating possible drug activity in a Kansas grocery parking lot, and was later killed after an armed standoff, authorities said Monday.

Hundreds gathered outside Topeka police headquarters with candles in memory of Cpl. David Gogian and Officer Jeff Atherly. Members of the slain officers' families attended Monday night's vigil along with dozens of law enforcement officers in uniforms.

"You never really think you're going to lose friends. Everybody, I think, is still in shock," said Officer Kurtis VanDonge, a 31-year-old Army veteran who was in Atherly's training class and worked under Gogian.

A bagpiper played "Amazing Grace," and a minister read from the opening chapter of the Gospel of John that includes the verse: "The light shines in the darkness, and the darkness has not overcome it."

The man who opened fire on the officers Sunday night was David Edward Tiscareno, 22, of Topeka, said Kyle Smith, deputy director of the Kansas Bureau of Investigation.

Shawnee County Sheriff Herman Jones said Tiscareno was seated behind the driver's seat of a car stopped in the parking lot of a Topeka grocery store when police ordered the occupants to get out. After shooting two of the three responding officers, Tiscareno got back into the car and drove from the scene. The third officer returned fire, Jones said.

"There were people inside the store," Jones said during a news conference earlier Monday. "There were residents who saw this going on. There were people driving. There was a multitude of witnesses there."

Gogian, 50, and Atherly, 29, were shot in the head, according to authorities.

Based on a tip, law enforcement officers found Tiscareno holed up early Monday at a house about a mile away. When negotiations broke down, Kansas Bureau of Investigation agents fired tear gas inside. Tiscareno then emerged from the home with a gun and officers opened fire, according to the Shawnee County sheriff's office, which is leading the investigation.

Smith said investigators believe Tiscareno fired a single shot outside the home before authorities returned fire. Tiscareno was declared dead at a hospital.

Police Chief Ronald Miller said the 280 officers on his Topeka force are "numb." The shootings in the Kansas capital followed last week's massacre at a Newtown, Conn., elementary school. Closer to Topeka, earlier this month, Kansas City Chiefs linebacker Jovan Belcher killed his girlfriend, then drove to the team's practice site and committed suicide.

"There's a lot of people in this country that are grieving some of the things that have happened over the last several days, and we're just counted among that group," Miller said.

Gov. Sam Brownback and Attorney General Derek Schmidt participated in the vigil. Miller called Gogian and Atherly "first-rate police officers" and added: "We can't make sense of what has happened."

Online court records show Tiscareno was charged in September 2008 with one count of theft of less than $1,000. He pleaded no contest in April 2009 and was sentenced to 12 months of supervised probation. His probation was revoked in January 2010.

In March 2011, he was charged with criminal use of weapons and was sentenced that September to 12 months of unsupervised probation, also after pleading no contest. A notation in court records available online said, "This defendant should be advised against carrying a firearm."

Court records also show that in February 2011, Tiscareno was evicted from an east Topeka apartment for failing to pay the rent and that a local hospital sued him for failing to pay almost $9,100 in bills.

Jones said he wasn't sure whether there was a drug deal or drug use going on in the store parking lot; the person who called authorities reported drug activity.

VanDonge said the officers' deaths were a shock because a situation like the one they were in wouldn't typically result in violence.

The third officer at the scene was not hurt and has been placed on administrative leave with pay, which is standard procedure when an officer is involved in a shooting, authorities said.

The other people who were in the car when the shootings happened have been questioned. Authorities would not say whether they might face charges.

Miller said he had hoped that Tiscareno would be arrested so that he could stand trial.

"One of the things that concerns me is that we don't really know why he did what he did," Miller said.

The last time a Topeka officer was killed in the line of duty was 2000, and it's been longer than that ? since 1995 ? that one was fatally shot on the job.

Gogian had been a Topeka police officer since September 2004 and had an adult son who's also a Topeka officer. Atherly joined the department in April 2011.

Gogian was a retired Kansas Air National Guard sergeant and a former Marine Corp reservist. A biography prepared for his retirement and released by guard officials said Gogian and his wife had four children.

Janet Rice, who works in the Topeka schools' business office, said Atherly was patient and kind. She's a volunteer ambassador for the department and rode a shift with Atherly in May, after graduating from a 13-week citizens' course on policing.

"He was always polite, even though there were people calling him all kinds of bad names," Rice recalled after the vigil. "This is a huge loss."
